Claims about its powers range from attuning one’s mind with God or even telepathic communication. Let’s look at what the science has to say about the powers of the pineal gland. THE PINEAL GLAND
The first thing to look at is this whole “third eye” thing. Is it actually that similar to the eyes we see with? Or could it be some vestige of an eye that was previously there? In other animals, it is even directly connected to the eye.
Many vertebrates across the mammal, reptile, and fish kingdoms have evolved a pineal gland. Others have evolved not to have one. In the case of the New Zealand reptile the tuatara, the pineal gland is directly connected to a literal third eye! Technically, the parietal eye is the correct gland to call a third eye. This gland appears on the tuatara’s head with a lens, retina, and cornea of its own. This connects to the pineal gland, as do the glowing third eyes of various deep-water fish. From an evolutionary standpoint, the pineal gland is clearly important to circadian rhythm, hormonal regulation, and things like that. But what else does it do for humans? The pineal gland receives blood flow directly from the rest of the body through the posterior cerebral artery. Since the blood is not processed through the blood-brain barrier, psychoactive chemicals hit the pineal gland much quicker. Melatonin performs several functions in the central nervous system, the most important modulating sleep patterns. Melatonin release is inhibited by light and stimulated by darkness. Light sensory nerve cells in the retina detect blue light and send a signal to the suprachiasmatic nucleus.

The suprachiasmatic nucleus – SCN is a tiny region of the brain in the hypothalamus (brain), situated directly above the optic chiasm which is connected to the eye (retinal nerves) contains only 20,000 neurons. It is responsible for controlling circadian rhythms over the 24-hour cycle. The neuronal and hormonal activity of the SCN regulates many different body functions. The SCN interacts with other regions of the brain and has several cell types that produce different peptides such as the hormone vasopressin, vasoactive intestinal peptide and neurotransmitters.
It is important to synchronize the SCN to the day-night cycle. Nerve fibers then relay retinal activation (to blue light) from the SCN to the paraventricular nuclei or PVN.
The paraventricular nucleus is a nucleus in the hypothalamus (brain) where its neurons are activated by physiological changes including stress. The PVN neurons project directly the pituitary where oxytocin is released into the blood. Oxytocin has hormonal) action, and acts on the brain. There is a oxytocin receptor that requires magnesium and cholesterol to work properly. Oxytocin’s has a role in behaviors, including orgasm, social recognition, pair bonding, anxiety, and maternal behaviors. Other neurons in the hypothalamus – pituitary-system are involved directly in the regulation of appetite and autonomic functions in the brainstem and spinal cord.
STUDY – RODENTS
Studies on rodents find that pineal gland probably influences the secretion of the sex hormones, hair follicle-stimulating hormone, and luteinizing sex (testosterone) hormones.
Studies on rats suggest that the pineal gland probably interacts with recreational drugs and that its hormone melatonin protects against neurodegeneration. Recreational drugs which interfere with the activity of the pineal, like cocaine and other hard drugs is conjectured to cause significant misbehavior.
Studies in mice suggest that the pineal-derived melatonin regulates new bone deposition, which is consistent with the observed bone stimulating growth of CBD, the second most common phytocannabinoid of marijuana. Pineal-derived melatonin mediated action on the bone cells through MT2 receptors. This pathway could be a potential new target for osteoporosis treatment.
Pinoline has long been claimed to be produced in the pineal gland during the metabolism of melatonin. Pinoline is a free radical scavenger, an antioxidant.
One of pinoline’s pharmacological properties is its ability to promote neurogenesis in vitro; even at trace concentrations in common with several cannabinoids like THC and CBD. Further study is required to firmly establish the exact mechanism of synergy between cannabis components, but the scientifically grounded implications is there in spades.
Aluminium toxicity causes lipid peroxidation damage in the brain. A recent review of studies shows both pinoline and melatonin to be effective at reducing the lipid peroxidation. If cannabinoids can be shown to elevated these brain chemicals to optimal levels, then their efficacy is obvious.
Lipopolysaccharide produced by bacterial infection stimulates the production of free radicals, which cause lipid peroxidation. Melatonin and pinoline were demonstrated in the ability to reduce lipid peroxidation damage. Pinoline was also shown to be better than vitamin E in reducing lipopolysaccharide activity in the retina, yet another overlap with cannabis, which is well known for its ability to combat glaucoma.
Lipopolysaccharides composed of a fat and sugar(s). and are found on the membrane of bacteria, which cause strong immune responses. These rogue fat-sugars may hold a key to inflammation and autoimmune disorders. Cannabinoids have the ability to control or reduce inflammation in many disorders, and interfere with the reproduction of common pathogenic bacteria (gram-negative). This is yet another avenue where cannabinoids and the “melatonin system” may work together to combat autoimmune disorders.
